چکیده

We report here a useful method for the isolation and culti vation of human tumor cells in vitro from human tumors grown in nude mice. A rabbit was immunized with spleen cells ob tained from adult nude mice. The rabbit antiserum in the presence of complement effectively killed cultured cells derived from various mouse tissues, but it was not cytotoxic to cultured cells from human tissues including tumors. When mixed cul tures consisting of human tumor cells and nude mouse fibroblasts were treated with the antiserum and complement, the nude mouse fibroblasts were completely removed from the cultures, and the human tumor cells could be propagated without noticeable changes in morphological features. Primary cultures of heterotransplanted human tumors grown in nude mice were also successfully treated, resulting in the ultimate elimination of fibroblastic cells derived from the stroma of the tumor. The functional properties of the tumor cells (production of human chorionic gonadotropin by choriocarcinoma cells and production of carcinoembryonic antigen by pancreas carcinoma cells) were also maintained after the antiserum treatment.

Xenografts of Human Bladder Cancer in Immune-Deprived Mice1

Human bladder cancer from cystoscopic biopsies and from established cell lines was transplanted into mice that were immune suppressed by thymectomy plus sequential treatment with 1-ß-0-arabinofuranosylcytosine and whole-body irradia tion. Each of four established human bladder cancer cell lines generated transplantable tumors in these mice, and some of the mice developed pulmonary métastases....

Evaluation of Growth and Histology of Human Tumor Xenografts Implanted under the Renal Capsule of Immunocompetent and Immunodeficient Mice1

Fresh surgical expiants of human carcinomas were implanted as first transplant generation xenografts under the kidney cap sule of mice. Immunocompetent and immune-deficient mice were compared for their ability to support the persistence and growth of these xenografts. Consistent growth of tumor xenografts could not be demonstrated following implantation under the kidney capsule of immunocompete...

Tumorigenicity of Esophageal Cancer Stem Cells (ECSCs) in nude mouse xenograft model

Background and objectives: Modeling cancer in vivo is a very important tool to investigate cancer pathogenesis and molecular mechanisms involved in cancer progression. Laboratory mice are the most common animal used for rebuilding human cancer in vivo. Cancer stem cells (CSCs) are the main reason of failure in cancer therapy because of tumor relapse and metastasis. Isolation of cancer stem cell...

Comparative Morphometric Study of Tumor Vasculature in Human Squamous Cell Carcinomas and Their Xenotransplants in Athymic Nude Mice1

When human tumor xenotransplants into nude mice are used as experimental models, it is important to know whether their microvascular anatomy is rather host or tumor specific. Therefore a morphometric comparison of the vascular network in human squamous cell carcinomas and their xenotransplants was carried out. Biopsies were taken from surgical specimens of three squamous cell carcinomas of the ...
